Possible side effects

Drug interaction

Sildalis has the potential for significant drug interactions. The most critical interaction is with nitrates and nitric oxide donors, which is absolutely contraindicated. Concomitant use with alpha-blockers (e.g., doxazosin, tamsulosin) can potentiate blood-pressure-lowering effects and may lead to symptomatic hypotension. Caution is advised when co-administered with other antihypertensive agents. Sildalis is primarily metabolized by the CYP3A4 enzyme system. Therefore, potent CYP3A4 inhibitors such as ketoconazole, itraconazole, ritonavir, and clarithromycin can significantly increase the concentration of Sildalis and should be avoided or used with extreme caution and dose adjustment. Grapefruit juice may also increase plasma levels and should be avoided.

Missed dose

Sildalis is not intended for daily scheduled use; it is taken on an as-needed basis. Therefore, the concept of a “missed dose” does not apply. The medication should only be taken when planned sexual activity is anticipated. Do not take an extra tablet to make up for a missed opportunity.

Overdose

In cases of suspected overdose, standard supportive measures should be applied. Given the vasodilatory effects of Sildalis, the primary expected manifestation would be severe hypotension. Management of overdose should include cardiovascular support, including placing the patient in the supine position with the legs elevated. Active management of hypotension may require fluid administration. Hemodialysis is not expected to accelerate clearance due to high protein binding and extensive tissue distribution. Priapism is another potential consequence of overdose and requires immediate urological intervention to prevent permanent tissue damage. In case of overdose, seek immediate medical attention or contact a Poison Control Center.

Storage

Store Sildalis tablets at room temperature, between 15°C and 30°C (59°F and 86°F), in their original packaging to protect from light and moisture. Keep the container tightly closed. Always store all medications out of the reach of children and pets. Do not use medicine past the expiration date printed on the packaging. Properly discard any unused or expired medication through a medicine take-back program or by following FDA-approved disposal guidelines; do not flush down the toilet or pour into a drain.
